چکیده

T HE EFFECT OF ANDROGENIC STEROIDS on hemoglobin levels has usually been ascribed to an indirect effect of these hormones, possibly secondary to an effect upon erythropoietin production or activation.1’2 Earlier studies of the in vitro effect of androgens on bone marrow cells provided conflicting evidence for a direct effect upon erythroid cell proliferation or hemoglobin synthesis. Reisner3 presented evidence for an in vitro effect upon iron incorporation by canine bone marrow cells whereas other workers were unable to confirm these results.4 In earlier studies, we could not document an in vitro effect of testosterone on hemoglobin synthesis, nucleic acid synthesis, or erythroid cell proliferation. Recently Granick and Kappas and their coworkers57 have published a series of papers in which they report a potent stimulation of heme synthesis in embryonic chick liver and erythropoietic tissues using certain metabolic derivatives of androgenic steroids. We have now demonstrated a similar in vitro effect of 5 fl-H metabolites of testosterone in adult human erythropoietic tissue. These data add support to the concept that androgenic hormones or their metabolic products may act directly upon human red cell precursors.

The influence of steroid hormone metabolites on the in vitro development of erythroid colonies derived from human bone marrow

Certain C19 and C21 steroid metabolites, when incubated with normal human bone marrow cells in culture, increased the number of erythroid colonies in the presence of erythropoietin. Among a number of pairs of C5 epimeric steroids tested, most 5beta (A:B cis) steroids stimulated the growth of both early erythroid progenitor cells (BFU-E) and late erythroid progenitor cells (CFU-E), whereas only ...

Enhancement of Heme and Globin Synthesis in Cultured Human Marrow by Certain 5β-h Steroid Metabolites

These studies demonstrate that certain sex steroid metabolites are capable of significantly stimulating the synthesis of both heme and globin in cultured human bone marrow cells. These compounds, which are maximally effective at a concentration of 3 x 10(-8)M, are steroids of the C(19) and C(21) neutral type; share a common 5beta-H (A:B cis) configuration; and are derived from the in vivo biotr...

Erythropoietic activity of steroid metabolites in mice.

The per cent Fe(59) incorporation into circulating erythrocytes as an indicator for new hemoglobin production was used in the polycythemic exhypoxic mouse system to study the effects of certain steroid hormone metabolites on erythropiesis. Enhanced Fe(59) incorporation was observed after the administration of several metabolites with a 5beta-H configuration, while those with a 5alpha-H configur...

In vitro Effect of Pomalidomide on Bone Marrow Mononuclear Cells from Multiple Myeloma Patients

Background: Many features of anticancer drugs, including cytotoxicity and/or cytokine induction, are studied using cell lines orhuman blood leukocytes. However, in a disease such as multiple myeloma, most cancerous cells are resided within bone marrowmononuclear cells. In the present study, we investigated the effect of pomalidomide on apoptosis and IL-2 production of bonemarrow mononuclear cel...
